Effects of red wine on ochratoxin A toxicity in intestinal Caco-2/TC7 cells. Abstract Ochratoxin A (OTA) is found in a variety of foods and beverages, including red wine. OTA was reported to be nephrotoxic, immunotoxic, hepatotoxic and a potential carcinogen, with yet uncharacterized mechanisms. Consumption of contaminated wines might contribute up to 13% of OTA daily human intake. Potentially chronic exposure has therefore raised public health concern. OTA toxicity in the presence of de-alcoholated red wine was investigated in human intestinal Caco-2/TC7 cells, differentiated on filter supports, by measuring tight junction (TJ) permeability, morphological alterations of TJ proteins and occurrence of apoptosis. Cells were treated with OTA, in the presence of de-alcoholated red wine, for 48h and the ability to recover from the effects of OTA was evaluated after 24h in complete medium. OTA treatment increased TJ permeability and caused intracellular redistribution of claudin-4. However, cells were able to restore permeability and correct localization of claudin-4 following 24h recovery. Conversely, in the presence of red wine, OTA produced faster and irreversible increase in TJ permeability, intracellular delocalization of claudin-4 and extensive apoptosis. Our results point at a possible synergy between OTA and some red wine components, such as polyphenols, in the induction of apoptotic cell death.

Canopy shade causes a rapid and transient arrest in leaf development through auxin-induced cytokinin oxidase activity. A plant grown under canopies perceives the reduction in the ratio of red (R) to far-red (FR) light as a warning of competitio n, and enhances elongation growth in an attempt to overgrow its neighbors. Here, we report that the same low R/FR signal that induces hypocotyl elongation also triggers a rapid arrest of leaf primordium growth, ensuring that plant resources are redir ected into extension growth. The growth arrest induced by low R/FR depends on auxin-induced cytokinin breakdown in incipient vein cells of developing primordia, thus demonstrating the existence of a previously unrecognized regulatory circuit underlyi ng plant response to canopy shade. Genes whose products function together are likely under a common regulatory system (e.g., signal transduction pathways, sets of regulatory proteins) such that they are expressed in a coordinated manner. This property has been frequently used in the analysis of genome-wide expression data, as the experimental observation that a group of genes is co-expressed frequently implies that the genes share a common regulato ry mechanism. The authors have investigated the situation in which dissimilarity in gene-expression time profiles may still r esult from the presence of the same regulatory signal, as in the case of common transcription factors. To this aim, a dynamic model that takes into account the effect of specific mRNA degradation on the shape of gene-expression time series has been d eveloped, and the concept of 'dynamically co-regulated' genes has accordingly been introduced as the goodness-of-fit to such a model (called dynamic R2). The statistical analysis of dynamic R2 over a number of different experimental data sets and org anisms shows that the presence of dynamically co-regulated genes is by far more significant than that expected from the rando mised data. Furthermore, as an example of the usefulness of the proposed method, genome-wide yeast measurements such as cell- cycle time series and transcription factors targets data, were used to prove that dynamic co-regulation is statistically rela ted to the presence of common transcription factor(s). This latter property is very useful when trying to infer computational indications of co-regulation for not-yet annotated genes that do not display a co-expression pattern. Functional analysis of transcription factors by microparticle bombardment. This chapter describes a method for rapid gene expression assays in Arabidopsis. Three distinct plasmids are codelivered into leaves by microparticle bombarment. The first one carries a transacting factor gene driven by the cauliflower mosaic virus ( CaMV) 35S promoter. The second plasmid includes a reporter luciferase (LUC) gene transcribed from a promoter containing the T F binding site(s). The third plasmid contains a reference beta-glucuronidase (GUS) gene transcribed from the 35S-CaMV promote r, which is used to normalize data derived from independent experiments. This method could be applied to study systematically the functional properties of Arabidopsis transcription factors as well as to identify promoters and control region(s) regula ted by specific transcription factors. Metal chelates of 2-hydroxy-4-methylthiobutanoic acid in animal feeding. Part 2: Further characterizations, in vitro and in vivo investigations.

AbstractThe alpha-hydroxyacid 2-hydroxy-4-methylthiobutanoic acid (the so-called methionine hydroxy-analogue, MHA), largely used in animal nutrition as a source of methionine, forms stable metal chelates with divalent metals of formula [{CH(3)SCH(2) CH(2)CH(OH)COO}(2)M].nH(2)O. Protonation and iron(III) and copper(II) complex formation constants have been determined by potentiometry at 25 degrees C. Distribution diagrams show that no free Fe(3+) cations are present in solution at pH>2.5. ESI-MS (Electron-Spray Ionization Mass Spectrometry) investigations carried out both on iron and zinc complexes in solution have evidenced various species with different MHA/metal ratios. In vivo trials were carried out with rats. After receiving a zinc-deficient diet for 3 weeks, animals were fed the same diet added with zinc sulfate or zinc/MHA chelate; the zinc content of faeces was higher (+45%; P<0.05) in sulfate fed rats, whereas zinc retention was higher (+61%; P<0.05) in the Zn/MHA diet. Experiments in vitro with human intestinal Caco-2 cells indicated that the MHA/Fe chelate was taken up by the cells without any apparent toxic effect. The iron uptake was higher than that of iron nitrilotriacetate (Fe(3+)NTA), an effective chelate for delivering iron to milk diets. In conclusion, these data indicate that the use of MHA chelates could be a valuable tool to increase bioavailability of trace minerals and reduce the environmental impact of animal manure.

The Caco-2 cell line as a model of the intestinal barrier: influence of cell and culture-related factors on Caco-2 cell functional characteristics.

Abstract The human intestinal Caco-2 cell line has been extensively used over the last twenty years as a model of the intestinal barrier. The parental cell line, originally obtained from a human colon adenocarcinoma, undergoes in culture a process of spontaneous differentiation that leads to the formation of a monolayer of cells, expressing several morphological and functional characteristics of the mature enterocyte. Culture-related conditions were shown to influence the expression of these characteristics, in part due to the intrinsic heterogeneity of the parental cell line, leading to selection of sub-populations of cells becoming prominent in the culture. In addition, several clonal cell lines have been isolated from the parental line, exhibiting in general a more homogeneous expression of differentiation traits, while not always expressing all characteristics of the parental line. Culture-related conditions, as well as the different Caco-2 cell lines utilized in different laboratories, often make it extremely difficult to compare results in the literature. This review is aimed at summarizing recent, or previously unreviewed, data from the literature on the effects of culture-related factors and the influence of line sub-types (parental vs. different clonal lines) on the expression of differentiation traits important for the use of Caco-2 cells as a model of the absorptive and defensive properties of the intestinal mucosa. Since the use of Caco-2 cells has grown exponentially in recent years, it is particularly important to highlight these methodological aspects in order to promote the standardization and optimisation of this intestinal model.

An inter-laboratory study to evaluate the effects of medium composition on the differentiation and barrier function of Caco-2 cell lines.

Abstract Differentiated human intestinal Caco-2 cells are frequently used in toxicology and pharmacology as in vitro models for studies on intestinal barrier functions. Since several discrepancies exist among the different lines and clones of Caco-2 cells, comparison of the results obtained and optimisation of models for use for regulatory purposes are particularly difficult, especially with respect to culture conditions and morphological and biochemical parameters. An inter-laboratory study has been performed on the parental cell line and on three clonal Caco-2 cell lines, with the aim of standardising the culture conditions and identifying the best cell line with respect to parameters relevant to barrier integrity, namely, trans-epithelial electrical resistance (TEER) and mannitol passage, and of epithelial differentiation (alkaline phosphatase activity). Comparison of the cell lines maintained in traditional serum-supplemented culture medium or in defined medium, containing insulin, transferrin, selenium and lipids, showed that parameter performance was better and more reproducible with the traditional medium. The maintenance of the cell lines for 15 days in culture was found to be sufficient for the development of barrier properties, but not for full epithelial differentiation. Caco-2/TC7 cells performed better than the other three cell lines, both in terms of reproducibility and performance, exhibiting low TEER and mannitol passage, and high alkaline phosphatase activity.

A dynamic balance between gene activation and repression regulates the shade avoidance response in Arabidopsis. Plants grown under dense canopies perceive through the phytochrome system a reduction in the ratio of red to far-red light as a warning of competition, and this triggers a series of morphological changes to avoid shade. Several phytochrome signaling intermediates acting as positive regulators of accelerated elongation growth and induction of flowering in shade avoidance ha ve been identified. Here we report that a negative regulatory mechanism ensures that in the presence of far-red-rich light an exaggerated plant response does not occur. Strikingly, this unpredicted negative regulatory mechanism is centrally involved in the attenuation of virtually all plant responses to canopy shade. PMID: 16322556 [PubMed - indexed for MEDLINE]
